De Primetime Emmy Award voor beste komedieserie is een jaarlijkse Amerikaanse televisieprijs die gegeven wordt aan de beste komedieserie van dat jaar. Van 1960 tot 1964 werd deze categorie gecombineerd met komediespecials (programma's die één keer verschenen).
The Flintstones en Family Guy zijn de enige geanimeerde sitcoms die ooit genomineerd werden voor de prijs. In 2019 schreef Fleabag (Verenigd Koninkrijk) geschiedenis door als eerste buitenlands programma de prijs te winnen.
De drie grote zenders (ABC, CBS en NBC) zijn de grote slokoppen en wonnen de meeste prijzen.
